rooting vs applying a custom rom ?

Hello all and pardon my ignorance,

I have nexus one in the way and I'm trying to learn as much as possible,I do understand the benefit and risk of rooting the device my questions is do I have to have it root it before applying a custom rom ? example "cyanogen's v5.0 Beta 3 ROM"

Is Superboot another way to have the device root it ?

Basically installed a cook rom and rooted is included.
