Bit of n00b here, how can I access the file?

I assume its on the internal memory, and this doesn't show in windows when I connect the phone.

Do I extract the file from the rom zip (on the pc) modify then update with clockwork?

Use android commander to browse the files on the phone, pull the build.prop then push it back over.
